The best in hat-painting, click-man-and-kill him, and team based objective gameplay.

The Classes - How the game divides the classes up are in to Offense, Defense, and Support. These mean next to nothing. A Pyro can play defensively, a Demoman is ridiculous on offense. Only labeling them for the sake of posterity.

"Offense":
Scout
If you enjoy twitch shooters (Unreal Tournament) Scouts your bro. Focusing on speed, some small acrobatics(double/triple*/QUAD* jumps,) and meatshots with a variety of Scatterguns.

*Only if using weapons that otherwise have negatives attached to them.

Soldier
Rockets. Rocket Jumping. Rocket Juggling. Crit Rockets. Fast Rockets. Not-quite-as-fast Rockets. The Soldier is an offensive powerhouse. A slow walking speed, but with Rocket Jumping technique, the Soldier can get to the frontlines just as quickly as the Scout. At the sacrifice of a bit, or most, of his health though.

Pyro
Harassments the name, Ambushings the game. The Pyro isn't going to win a straight up fight most of the time. However, the disruption you can do as a Pyro is almost unmatched. He also boasts the ability to reflect most projectiles back at the person they came from, or to another target entirely, using up a chunk of his flamethrowers ammo to perform a blast of compressed air. This also works on players, and can easily break up a Medic from his Patient.

"Defense":
Demoman
The ability to perform some serious traps and area denial, as well as having an unmatched damage potential causes the Demoman to be one of the strongest classes in the game in the right hands. Recently he doubles as a 10th class, dubbed the Demoknight, as Valve has given the Demoman weapons that forgo the normal explosives and make him in to a dangerous melee brawler.

Heavy
TF2 Training Wheels. Not much you can say about the Heavy. He's big. He's slow. He's sturdy. He has a gun that chews through people pretty easily. If you're seen by a Heavy in mid-close range, you're already dead. Though he is probably the most MEME-orable(heh, get it?) portion of the game.

Engineer
Worthless by himself. Invaluable to a team. Good Engineers are absurdly rare. No doubt, his gameplay pretty much enforces laziness. Building Sentry Guns to defend areas, teleporters to move teammates around the map faster, and dispensers to resupply and heal teammates, an Engineer can make a break a victory. Can opt to take a smaller sentry gun that cannot be upgraded for a more offensive approach.

"Support":
Medic
Similar to the Engineer in terms of how they stack up on a scale of power. A Medic is more of a tool than anything else. He heals other players, keeping them up so that the offensive push/defensive hold can continue to do what they're doing. After healing enough, the Medic can deploy what they call an Ubercharge. The actual effect of this charge depends on which Medigun you're currently using. As of now, there are three. The default will give 8 seconds of invulnerability to whoever you're healing. The Kritzkrieg will give 8 seconds of guaranteed critical hits to teammates, increasing their damage output to ludicrous levels. The Quick Fix, again for 8 seconds, will increase the rate of the healing by 300% and defend against knockbacks of any kind. If TF2 had Team Captains, the Medic would definitely fit the role best, being a linchpin member to any team.

Sniper
Pretty much what you'd expect. Hit guys in the head, get points. Among the most boring of classes, it's amazing people actually play this one.

Spy
To sum up the Spy, you have to look at it from the perspective of someone playing against it. That is to say.. in the hands of a pro, a murder machine you cannot shake - In the hands of a scrub, easy points for you. The Spy can cloak and/or disguise himself as another class, both friendly and enemy. He can instantly kill any other player if he scores a successful backstab. He can disable Engineer buildings. He can shoot you with his revolver for a ton of damage. But he's incredibly frail. The glassiest of the cannons in this game.

Meet The Medic, it owns. Best video to date. Can't wait for Meet the Pyro, then the movie(Come on Valve, you know you want to make one.)
